This video will show you how to begin a day with Duopa. You’ll see how to set up the pump, how to give the different doses, and how to stop and disconnect the pump when needed.

About Duopa
TEXT ON-SCREEN and narrator:
Duopa (carbidopa and levodopa) is a prescription medicine used for treatment of advanced Parkinson’s disease. Duopa contains 2 medicines, carbidopa and levodopa.
The Duopa system uses a tube to administer (carbidopa and levodopa) enteral suspension directly to your intestine. A cassette containing the medication is attached to a portable pump. The pump connects to the stomach tube used to deliver the medication.

Narrator:
Take one Duopa cassette out of the refrigerator and out of the carton. Return the carton with remaining cassettes to the refrigerator. Do not use the cassette if the expiration date has passed or the cassette is damaged or empty. Let the cassette sit out at room temperature for 20 minutes before using.
Each cassette can be used for up to 16 hours after removal from the refrigerator.

WARNING! Attach the Duopa cassette correctly. A detached or incorrectly attached cassette could cause a problem with getting Duopa.
Narrator:
Now, let’s start by connecting the pump.
Remove the cassette tube from the secured slot in the clip. Then, pull the clip from the cassette to slide it off the cassette top.
Hold the cassette with the tube pointing down, and hold the pump with the latch facing up. Insert the cassette hooks into the hinge pins at the base of the pump.
Now, hold the pump and cassette upright against a flat surface and press down on the pump until the cassette fits tightly against it. Use a coin to twist the latch counterclockwise until latch lines up straight with the arrow.
Be sure to attach the Duopa cassette correctly. A detached or incorrectly attached cassette could cause a problem with getting Duopa.

Save the red cap for use when you throw away the cassette
WARNING! Do not connect the red cap to stomach tube. It will block Duopa flow
Connect tubing
WARNING! Do not twist the stomach tube
Do not connect to the angled connector. Your angled connector may be white, blue, or violet.
Narrator:
Remove the red cap on the end of the cassette tube. Save the red cap for use when you throw away the cassette.
Do NOT connect the red cap to the stomach tube, as this will block Duopa flow.
Now connect the stomach tube to the cassette tube. Start by holding the stomach tube steady and twisting the white cap off the straight green connector. Do not twist the stomach tube.
Next, connect the cassette tube to the end of the straight green connector. Do not connect it to the angled connector. Your angled connector may be white, blue, or violet.

Turn on the Pump
Press and hold ON/OFF button
Wait 30 seconds
Narrator:
To turn on the pump, press and hold the ON/OFF button until the display turns on. Wait about 30 seconds for the pump to review settings. The pump is ready when the display reads “STOPPED”. The pump is now on but not delivering medication.

Inspect tubing
Narrator:
Make sure the tubing is not twisted or kinked, and the clamps are open. Straighten kinks and/or open clamps if necessary.

Press and hold STOP/START button
Wait 15 seconds
Narrator:
Next, press and hold the STOP/START button until you see 3 dashes appear and disappear from the display. Wait 15 seconds until the word “RUN” appears. The pump is now running.
If the pump will not start, a message should appear on the display. Refer to the Alarms and Messages section in the Patient Instructions for Use.

Give the Morning Dose
Press MORNING DOSE button
Press MORNING DOSE button again
It will take between 10-30 minutes to deliver your morning dose.
If the morning dose is not delivered, enough time may not have passed since your last morning dose. You may need to wait longer.
Narrator:
Press the MORNING DOSE button once. The display will read “Morning Dose” along with showing the dose amount your healthcare provider prescribed for you. Press the MORNING DOSE button again, and a countdown will begin on the display. If the Morning Dose is not delivered after completing this step, enough time may not have passed since your last Morning Dose for another to be given. You may need to wait longer.

Continuous Rate will automatically start
Narrator:
After the Morning Dose is complete, the display will read “RUN”, and the pump will automatically start the Continuous Rate.
Put the pump into the carrying bag.

Press and hold STOP/START button
Narrator:
To stop the Continuous Rate, press and hold the STOP/START button until 3 dashes appear and disappear from the display.
Confirm that “STOPPED” appears on the display.

Change the Morning Dose
WARNING! Do not use the PRIME button. Priming is for use by your health care provider only.
Narrator:
Your healthcare provider may have set your pump to allow for dose adjustments to your Morning Dose and Continuous Rate.
Talk to your healthcare provider to decide when to change your Morning Dose and Continuous Rate. Do not change your medication or Extra Dose unless specifically instructed to by your healthcare provider.
WARNING: Do not use the PRIME button. Priming is for use by your health care provider only.

Make sure the pump is turned on, the line is clear of kinks, the clamps are open, and press the morning dose button once.
Narrator:
To begin changing the Morning Dose, make sure the pump is turned on and press the MORNING DOSE button once.
Your current Morning Dose should appear on the display. Press the up and down buttons to select your desired Morning Dose.

Press ENTER/CLEAR button
Narrator:
Next, press the ENTER/CLEAR button to store the Morning Dose. Confirm that you see the desired Morning Dose on the display. If not, repeat the previous steps.

Press MORNING DOSE button

If you see “Value not saved” on the display, press NEXT and repeat the preceding steps.
Narrator:
To deliver the Morning Dose, press the MORNING DOSE button once. The display will show a countdown of your Morning Dose. If you see “Value not saved” on the display, press NEXT and repeat the previous steps.
Your healthcare provider may not have set the pump to allow you to increase your Morning Dose which may result in “Value not saved”.

If you are unable to deliver a Morning Dose, it may be too soon since the last Morning Dose to deliver another.
Narrator:
After the Morning Dose finishes, the pump will automatically begin delivering the Continuous Rate and “RUN” will appear on the display.
If you are unable to deliver a Morning Dose, it may be too soon since the last Morning Dose to deliver another and you may need to wait longer. The time between Morning Doses is determined by your healthcare provider.
Do not attempt to adjust your medication without approval and training from your healthcare provider.

Change the Continuous Rate
Narrator:
To change the Continuous Rate, you need to stop the Continuous Rate first. To do this, press and hold the STOP/START button until 3 dashes appear and then disappear.
Check that the display reads STOPPED.

Press NEXT button twice
Narrator:
Next, press the NEXT button twice.

Press the up and down buttons to select desired Continuous Rate
Narrator:
Check for the Continuous Rate to appear on the display and press the up and down buttons to select the desired Continuous Rate.

Press ENTER/CLEAR button and confirm the desired Continuous Rate on the display
Narrator:
Press the “ENTER/CLEAR” button to store the Continuous Rate and confirm that you see the desired Continuous Rate on the display. If not, repeat the previous steps.

To start the pump
Press STOP/START button until 3 dashes appear and then disappear
Narrator:
To restart the pump, press and hold the STOP/START button until 3 dashes appear and then disappear. Wait approximately 15 seconds for the pump to start running.

If you see “Value not saved” on the display, press NEXT and repeat the preceding steps.
Narrator:
If “Value not saved” appears on the display when you start the pump, press the NEXT button and repeat the previous steps.
The display will show “RUN” and the pump is now running.

Give an Extra Dose
The time between Extra Doses is determined by your health care provider.
Narrator:
Your doctor may program your pump so you can give yourself Extra Doses as needed during the day. The time between Extra Doses is determined by your health care provider.

If the Extra Dose is not delivered, it may be too soon since the last Extra Dose to deliver another.
You may need to wait some time before delivering an Extra Dose.
Narrator:
To give yourself an Extra Dose, first ensure that the display of the pump reads “RUN”. Then press the EXTRA DOSE button once. Listen for 2 beeps. When the display reads “DOSE”, the pump is delivering the Extra Dose. When it’s finished, the display will read “RUN” and the Continuous Rate will resume.
If the Extra Dose is not delivered, it may be too soon since the last Extra Dose to deliver another. You may need to wait some time before delivering an Extra Dose.

Temporarily Disconnecting Pump or Changing Cassette
Narrator:
During the day, you may need to disconnect your pump to change your cassette or to take a shower, bath, or swim. No oral medication is needed if you stop your pump for less than 2 hours, but you may be advised by your physician to take an Extra Dose of Duopa before disconnecting. If you need to disconnect from your pump for more than 2 hours during the day, consult your physician. To change your cassette or disconnect your pump, follow these instructions:

Press and hold STOP/START button
Press and hold ON/OFF button
Narrator:
If changing your cassette, take the new cassette out of the refrigerator and let it stand at room temperature for 20 minutes before using, as you did in your morning procedure.
Now, remove the pump from the bag and stop the dose by pressing and holding the STOP/START button until 3 dashes appear and disappear. Check to see that the display reads “STOPPED”. Then, turn the pump off by pressing and holding the ON/OFF button until 3 sets of dots appear and disappear and the display turns off.

Narrator:
Next, clamp the cassette tube.
Gently twist the cassette tube (not the stomach tube) to disconnect it from the straight green connector on the stomach tube. Do not twist the stomach tube. Put the red cap back on the cassette tube.
At this point, you can continue with changing your cassette or bathing. Remember, do not get the pump wet. It is not waterproof.

Reconnect the Pump
Follow the same steps for connecting the cassette in the morning procedure.
Press and hold ON/OFF button
Check the tubing
Press and hold STOP/START button
Narrator:
When you are ready to reconnect the pump, connect the tubing as you did for the morning procedure. Press and hold the ON/OFF button until the display turns on. The pump will review its settings as before. When it is ready, the display will read “STOPPED”.
Check the tubing for kinks or closed clamps.
Then press and hold the STOP/START button until 3 dashes appear and disappear from the display. After about 15 seconds, the display will read “RUN”, meaning that the Continuous Rate has resumed.
USE
DUOPA (carbidopa and levodopa) enteral suspension is a prescription medicine used for treatment of advanced Parkinson’s disease. DUOPA contains two medicines: carbidopa and levodopa.
IMPORTANT SAFETY INFORMATION
What is the most important safety information I should know about DUOPA?
- Stomach and intestine (gastrointestinal) problems and problems from the procedure you will need to have to receive DUOPA (gastrointestinal procedure-related problems) may occur. Some of these problems may require surgery and may lead to death.
- Serious side effects may include: a blockage of your stomach or intestines (bezoar); stopping movement through intestines (ileus); drainage, redness, swelling, pain, feeling of warmth around the small hole in your stomach wall (stoma); bleeding from stomach ulcers or your intestines; inflammation of your pancreas (pancreatitis); infection in your lungs (pneumonia); air or gas in your abdominal cavity; skin infection around the intestinal tube, pocket of infection (abscess), or infection in your blood (sepsis) or abdominal cavity may occur after surgery; stomach pain, nausea, or vomiting. - Tell your healthcare provider right away if you have any of the following symptoms of stomach and intestine problems and gastrointestinal procedure-related problems: stomach (abdominal) pain; constipation that does not go away; nausea or vomiting; fever; blood in your stool; or a dark tarry stool.
Tell your healthcare provider about all the medicines you take, including prescription and over-the-counter medicines, vitamins, and herbal supplements. Using DUOPA with certain other medicines, including medications for high blood pressure, MAO inhibitors, antipsychotics, metoclopramide, isoniazid, and iron or vitamin supplements, may cause serious side effects. High-protein foods may affect how DUOPA works. Tell your healthcare provider if you change your diet.
DUOPA may cause serious side effects. Talk to your doctor before starting DUOPA and while on DUOPA if you have had or have any of these:
- Falling asleep during normal daily activities without warning. DUOPA may cause you to fall asleep while you are doing daily activities such as driving, which may result in an accident. This can happen as late as one year after starting DUOPA. Do not drive or operate machinery until you know how DUOPA affects you. Tell your healthcare provider if you take medicines that can make you sleepy, such as sleep medicines, antidepressants, or antipsychotics.
- Low blood pressure when you stand or sit up quickly. After you have been sitting or lying down, stand up slowly to help reduce dizziness, nausea, sweating, or fainting until you know how DUOPA affects you.
- Seeing, hearing, or feeling things that are not real (hallucinations).
- Unusual urges. Some people taking medicines for Parkinson’s disease, including DUOPA, have reported urges such as excessive gambling, compulsive eating, compulsive shopping, and increased sex drive.
- Depression and suicide. DUOPA can cause or worsen depression. Pay close attention to changes in your mood, behavior, thoughts, or feelings. Call your healthcare provider right away if you feel depressed or have thoughts of suicide.
- Uncontrolled sudden movements (dyskinesia). If you have new dyskinesia or your dyskinesia gets worse, tell your healthcare provider. This may be a sign that your dose of DUOPA or other Parkinson’s medicines may need to be adjusted.
- Progressive weakness or numbness or loss of sensation in the fingers or feet (neuropathy).
- Heart attack or other heart problems. Tell your healthcare provider if you have experienced increased blood pressure, a fast or irregular heartbeat, or chest pain.
- Abnormal blood tests. DUOPA may cause changes in certain blood tests, especially certain hormone and kidney function blood tests.
- Worsening of the increased pressure in your eyes (glaucoma). The pressure in your eyes should be checked after starting DUOPA.
Do not stop using DUOPA or change your dose unless you are told to do so by your healthcare provider. Tell your healthcare provider if you develop withdrawal symptoms such as fever, confusion, or severe muscle stiffness.
The most common side effects of DUOPA include: complications of tubing placement procedure, swelling of legs and feet, nausea, high blood pressure (hypertension), depression, and mouth and throat pain.
